目的观察氟化钠对小鼠睾丸间质瘤细胞(mLTC-1)中类固醇急性调节蛋白(StAR)和细胞色素P450胆固醇侧链裂解酶(P450scc)mRNA表达水平的影响。方法利用小鼠mLTC-1细胞为模型,用实时定量PCR法测定细胞中StAR和P450scc mRNA表达水平。采用放射免疫法测定细胞培养基上清中孕酮分泌量。结果与对照组比较,12、16和20μg/ml氟化钠浓度组的StAR和P450scc mRNA的表达量以及孕酮分泌量明显降低(P<0.05)。结论氟化钠可以抑制mLTC-1细胞StAR和P450scc mRNA的表达,进而影响mLTC-1细胞孕酮的合成。
Objective To observe the effects of sodium fluoride on the expression of steroid A regulatory protein (StAR) and cytochrome P450 cholesterol side chain lyase (P450scc) in mouse testicular stromal cells (mLTC-1). Methods The mouse mLTC-1 cells were used as a model to detect the expression of StAR and P450scc mRNA in real-time quantitative PCR. The progesterone secretion in the cell culture supernatant was determined by radioimmunoassay. Results Compared with the control group, the expression of StAR and P450scc mRNA and the secretion of progesterone in 12, 16 and 20μg / ml sodium fluoride group were significantly decreased (P <0.05). Conclusion Sodium fluoride can inhibit the expression of StAR and P450scc mRNA in mLTC-1 cells and further affect the progesterone synthesis in mLTC-1 cells.
