目的探讨大鼠肾缺血再灌注后不同时间点Smad7的表达,寻找临床预测急性肾功能衰竭的新指标及治疗的新手段。方法建立大鼠肾缺血再灌注模型,当再灌注达相应时间点时,采用全自动生化分析仪、免疫组化、HE染色分别测定或观察血肌酐、尿素氮、肾组织Smad7表达、肾组织病理改变。结果①肾功能改变:与假手术组比较,0h、4h、12h、24h时模型组BUN及Cr均高于假手术组(P<0.01)。②肾脏组织学改变:模型组的肾组织损伤较假手术组明显加重。③肾组织Smad7的表达:模型组与同一时间点假手术组比较,在0h时肾脏Smad7阳性表达两组表达无差异(P>0.05);在4h、12h、24h模型组肾脏Smad7阳性表达较假手术组均减少(P均<0.01)。结论随着肾功能逐渐恶化的同时Smad7表达不断减少。
Objective To investigate the expression of Smad7 at different time points after renal ischemia-reperfusion in rats and to find out a new index of clinical prediction of acute renal failure and a new therapeutic approach. Methods Rat renal ischemia-reperfusion model was established. When reperfusion reached the corresponding time point, the expression of Smad7 in serum creatinine, urea nitrogen and renal tissue were measured by immunohistochemistry and HE staining respectively. Pathological changes. Results ① Renal function changes: Compared with sham operation group, BUN and Cr in model group were significantly higher than those in sham operation group at 0h, 4h, 12h and 24h (P <0.01). ② renal histological changes: the model group of renal tissue injury was significantly worse than the sham group. (3) The expression of Smad7 in renal tissue: There was no difference in the expression of Smad7 between the model group and the sham operation group at the same time point (P> 0.05) at 0h; the positive expression of Smad7 in the model group at 4h, 12h and 24h The operation group was decreased (all P <0.01). Conclusion With the gradual deterioration of renal function, the expression of Smad7 is decreasing.
