目的 观察复方861对HSC-T6组织基质金属蛋白酶抑制因子1(TIMP1)基因表达的影响。方法复方861 0.25、0.5、1.0mg/ml等浓度作用于HSC-T6细胞48h,以逆转录定量PCR方法测定其对HSC-T6细胞TIMP1基因表达的影响。结果 复方861 0.25、0.5、1.0mg/ml等不同浓度作用后,HSC-T6细胞基因表达水平依次为2.50±0.71、0.50±0.01、0.11±0.03,与空白对照组门(3.78±0.67)比较,可明显抑制TIMP1基因表达水平(P<0.05或P<0.01)。结论 复方861抑制HSC-T6细胞TIMP1基因表达水平,从而促进胶原降解,可能是抗肝纤维化的作用机制之一。
Objective To observe the effect of compound 861 on the gene expression of tissue inhibitor of metalloproteinase-1 (TIMP1) in HSC-T6 tissue. Methods HSC-T6 cells were treated with different concentrations of 861, 0.25, 0.5 and 1.0 mg / ml for 48 h. The TIMP1 gene expression in HSC-T6 cells was determined by reverse transcription polymerase chain reaction (RT-PCR) Results Compared with blank control group (3.78 ± 0.67), the gene expression levels of HSC-T6 cells were 2.50 ± 0.71,0.50 ± 0.01,0.11 ± 0.03 after different concentrations of compound 861 0.25,0.5,1.0mg / ml, TIMP1 gene expression was significantly inhibited (P <0.05 or P <0.01). Conclusion Compound 861 can inhibit the expression of TIMP1 gene in HSC-T6 cells and promote collagen degradation, which may be one of the mechanisms of anti-liver fibrosis.
