目的:研究肿瘤坏死因子α(TNFα)对大鼠骨骼肌蛋白质降解具有促进作用的机制。方法:用TNFα处理在体、高体大鼠骨骼肌。结果:在体组TNFα处理后6h、12h比目鱼肌酪氨酸(Tyr)释放量非常显著地高于对照组(P<0.01);24h有所下降,但仍显著地高于对照组(P<0.05)。三时相点骨骼肌泛素(Ub)含量比对照组皆有显著地增加(P<0.01)。Tyr释放量、Ub含量变化趋势一致,相关非常显著(P<0.01)。结论:TNFα具有全身效应,能通过循环系统作用于骨骼肌,并通过增加Ub含量促进骨骼肌蛋白质降解。而TNFα处理后的离体骨骼肌Ub、Tyr释放量皆无显著变化,提示TNFα对Ub系统的作用可能不是直接的,可能还有机体其它因素的参与。
Objective: To study the mechanism of tumor necrosis factor α (TNFα) on the degradation of skeletal muscle protein in rats. Methods: In vivo and in vivo rat skeletal muscle were treated with TNFα. RESULTS: The release of tyrosine (Tyr) from the soleus muscle was significantly higher than that of the control group (P <0.01) at 6h and 6h after the TNFα treatment, but decreased significantly at 24h, but still significantly higher than that of the control group P <0.05). Compared with the control group, Ubiquitin (Ub) content increased significantly at the third hour (P <0.01). Tyr release, Ub content trend of the same, the correlation is very significant (P <0.01). CONCLUSIONS: TNFα has a systemic effect that acts on skeletal muscles through the circulatory system and promotes the degradation of skeletal muscle proteins by increasing Ub content. However, there was no significant change in Ub and Tyr release from ex vivo skeletal muscle after TNFα treatment, suggesting that the effect of TNFα on Ub system may not be direct and may involve other factors in the body.
