目的:研究猕猴桃多糖对615小鼠胃癌原位移植瘤的抑瘤效应及其与增殖细胞核抗原(PCNA)、p53表达的关系。方法:用OB吻合胶粘贴法建立615小鼠胃癌原位移植瘤模型。40只615小鼠随机分为5组:猕猴桃多糖高、中、低剂量组、5-FU对照组、模型对照组。计算抑瘤率、脾指数,并用免疫组织化学方法检测PCNA、p53的表达。结果:与模型对照组比较,用药各组平均瘤重明显下降(P<0.01);猕猴桃多糖中、高剂量组的脾指数明显升高(P<0.05,P<0.01);用药各组PCNA、p53的平均阳性指数有显著下调(P<0.01)。结论:猕猴桃多糖对615小鼠胃癌原位移植瘤具有抑制作用,且随着剂量的增加其抗肿瘤作用增强。其作用途径可能与提高小鼠的免疫功能及下凋PCNA、p53的表达有关。
Objective: To study the antitumor effect of Actinidia chinensis polysaccharides on orthotopic transplanted gastric cancer 615 mice and its relationship with the expression of proliferating cell nuclear antigen (PCNA) and p53. Methods: An in situ xenograft model of 615 mice with gastric cancer was established by OB anastomosis adhesive method. 40 615 mice were randomly divided into 5 groups: high, medium and low doses of kiwifruit, 5-FU control group and model control group. The tumor inhibition rate and spleen index were calculated, and the expression of PCNA and p53 were detected by immunohistochemistry. Results: Compared with the model control group, the average tumor weight in each group decreased significantly (P <0.01); the spleen index of high and medium dose groups of Kiwifruit increased significantly (P <0.05, P <0.01) The average positive index of p53 was significantly down-regulated (P <0.01). Conclusion: Actinidia chinensis polysaccharides can inhibit the growth of orthotopic transplanted gastric cancer 615 in 615 mice, and its anti-tumor effect is enhanced with the dose increasing. Its mechanism of action may be related to improving the immune function and the expression of PCNA and p53 in mice.
