目的:比较糖尿病性心肌病大鼠和中药复方糖心康治疗组大鼠心肌组织中基因表达的差异,并用Evegreen实时荧光RT-PCR技术定量检测膜联蛋白A4(annexin A4)在各组大鼠心肌组织的表达水平。方法:(1)差异表达基因的筛选;(2)荧光定量RT-PCR的检测。结果:本文仅对筛选出差异表达基因谱中的annexin A4 mRNA进行报告。糖尿病性心肌病模型组心肌组织基因annexin A4 mRNA表达下调,中药复方糖心康治疗组心肌组织基因annexin A4 mRNA表达上调。结论:annexin A4 mRNA在糖尿病状态下心肌组织也有表达且表达下调,而中药复方糖心康可使其上调,这也是中药复方糖心康对糖尿病性心肌组织损伤具有保护作用的机制之一。
OBJECTIVE: To compare the differences of gene expression in myocardium between diabetic rats with cardiomyopathy and traditional Chinese medicine Tangxinkang, and to detect the expression of annexin A4 by Evegreen real-time RT-PCR in rats Myocardial tissue expression levels. Methods: (1) Screening of differentially expressed genes; (2) Fluorescence quantitative RT-PCR. Results: Only Annexin A4 mRNA in differentially expressed genes was screened out. The expression of annexin A4 mRNA in myocardial tissue of diabetic model group was down-regulated, while the expression of annexin A4 mRNA in myocardial tissue of traditional Chinese medicine compound group was up-regulated. Conclusions: Annexin A4 mRNA is also expressed and down-regulated in diabetic rats, which is up-regulated by traditional Chinese medicine compound Tangxinkang. It is also one of the mechanisms that Tongxinkang has a protective effect on diabetic myocardial injury.
