目的探讨红色毛癣菌几丁质合酶(chitin synthase,CHS)m RNA在甲真菌病体外模型中的改变及其在致病中的作用。方法构建甲真菌病体外模型,随机分为实验组和对照组(未加甲板),分别于培养的第0,1,3,5,7,14和20天提取红色毛癣菌总RNA,采用实时荧光定量PCR检测不同时间点CHS m RNA的表达水平,并进行统计学分析。结果对照组CHS m RNA表达水平极低,实验组在红色毛癣菌感染甲板1d,3d,5d表达升高,7d表达达高峰,此后渐渐下降,但仍高于对照组。各时间点与对照组相比,除CHS 3d外,其余差异均有统计学意义(P<0.05)。结论 CHS m RNA在甲真菌病体外模型中有较高表达,在甲真菌病的致病中可能起重要作用。
Objective To investigate the changes of chitin synthase (CHS) m RNA in the in vitro model of onychomycosis and its role in pathogenicity. Methods The in vitro model of onychomycosis was constructed and randomly divided into experimental group and control group (without deck). The total RNA of Trichophyton rubrum was extracted on the 0, 1, 3, 5, 7, 14 and 20 days of culture, Real-time quantitative PCR was used to detect the expression of CH m RNA at different time points and statistical analysis was performed. Results The expression of CHS m RNA in the control group was extremely low. The expression of CHS m RNA in the experimental group increased on the 1d, 3d, 5d, and reached the peak on the 7th day after the infection of Trichophyton rubrum, and then decreased gradually, but still higher than that of the control group. Compared with the control group at all time points, except CHS 3d, the other differences were statistically significant (P <0.05). Conclusion CHS m RNA is highly expressed in the in vitro model of onychomycosis and may play an important role in the pathogenesis of onychomycosis.
