目的通过分析不同程度血糖状态与实验性糖尿病大鼠心肌胰岛素样生长因子1受体 mRNA(IGF-IR mRNA)表达数与心肌形态的关系,明确 IGF-1R mRNA 在糖尿病心肌病中的作用。方法制作糖尿病大鼠模型,于成模后3个月经光镜和透视电镜观察心肌的形态学改变。应用原位杂交技术对不同血糖状态下 IGF-1R mRNA 在心肌上的表达情况进行观察。结果 IGF-1RmRNA 在正常大鼠及糖尿病鼠(模型 B 组、C 组)心肌细胞膜和胞质中均有表达,其表达率分别为20%、30%和38%,且血糖及糖化血红蛋白 Al(HbA1c)水平与糖尿病大鼠心肌细胞膜及胞质 IGF-1RmRNA 表达数呈正相关,相关系数分别为0.869和0.865(均 P<0.01),表明与心肌形态结构改变相关。结论随着血糖水平升高,糖尿病大鼠心肌 IGF-1R mRNA 表达量增加,心肌形态结构改变加重,IGF-1R mRNA 表达量的变化很可能是糖尿病心肌病变发生、发展的重要因素。
OBJECTIVE: To determine the role of IGF-1R mRNA in diabetic cardiomyopathy by analyzing the relationship between the expression of insulin-like growth factor 1 receptor (IGF-IR mRNA) and myocardial morphology in different degree of blood glucose and experimental diabetic rats. Methods The model of diabetic rats was made. Morphological changes of myocardium were observed by light microscope and transmission electron microscope 3 months after the model was established. In situ hybridization was used to observe the expression of IGF-1R mRNA in myocardium under different blood glucose levels. Results IGF-1R mRNA was expressed in the membrane and cytoplasm of normal rats and diabetic rats (model group B and group C), the expression rates of which were 20%, 30% and 38%, respectively. The levels of blood glucose and glycosylated hemoglobin A1 ( HbA1c) was positively correlated with the expression of IGF-1R mRNA in cardiomyocytes and cytoplasm of diabetic rats, the correlation coefficient was 0.869 and 0.865 respectively (all P <0.01), which showed that the level of HbA1c was correlated with the change of myocardial morphology. Conclusion With the increase of blood glucose level, the expression of IGF-1R mRNA in myocardium of diabetic rats increases and the changes of myocardial morphology and structure change. The change of IGF-1R mRNA expression may be an important factor in the occurrence and development of diabetic cardiomyopathy.
