目的:通过探讨参附注射液对新生大鼠心力衰竭心肌细胞Ca2+-ATP酶,Ca2+-Mg2+-ATP酶、Na+-K+-ATP酶活性及细胞内Na+,Mg2+,K+,Ca2+浓度的影响,揭示参附注射液强心作用的机制。方法:通过胰蛋白酶消化法和差速贴壁法获得心肌细胞,经0.8%戊巴比妥钠作用5min后,分别给予1.5,3,6 mg.L-1(5,10,20 mL.L-1)3个不同浓度的参附注射液,作用1 h,检测心肌细胞Ca2+-ATP酶,Ca2+-Mg2+-ATP酶,Na+-K+-ATP酶的活性和细胞内Na+,Mg2+,K+,Ca2+的浓度。结果:参附注射液能增加心衰模型心肌细胞的搏动强度,提高Ca2+-ATP酶和Ca2+-Mg2+-ATP酶的活性,抑制Na+-K+-ATP酶的活性,降低细胞内K+,Ca2+的浓度,升高细胞内Na+,Mg2+的浓度。结论:参附注射液对戊巴比妥钠致心衰细胞有明显的保护作用,且作用的发挥与调节心肌细胞Ca2+-ATP酶,Ca2+-Mg2+-ATP酶,Na+-K+-ATP酶的活性和细胞内Na+,Mg2+,K+,Ca2+的浓度有关。
Objective: To investigate the effect of Shenfu injection on the activity of Ca2 + -ATPase, Ca2 + -Mg2 + -ATPase, Na + -K + -ATPase and the intracellular Na +, Mg2 +, K + and Ca2 + concentrations in cardiomyocytes of neonatal rats with heart failure Shen Fu injection cardiac effect of the mechanism. Methods: Cardiomyocytes were obtained by trypsin digestion and differential adherent method. Results: Shenfu injection can increase the pulsatility of cardiomyocytes in heart failure model, increase the activity of Ca2 + -ATPase and Ca2 + -Mg2 + -ATPase, inhibit the activity of Na + -K + -ATPase and decrease the concentration of K +, Ca2 + , Increase the concentration of intracellular Na +, Mg2 +. CONCLUSIONS: Shenfu injection has a significant protective effect on heart failure cells induced by pentobarbital sodium, and its function is related to the activity of Ca2 + -ATPase, Ca2 + -Mg2 + -ATPase and Na + -K + -ATPase in cardiomyocytes And intracellular Na +, Mg2 +, K +, Ca2 + concentration.
