目的 :探讨增殖性肾小球肾炎大鼠动物模型中系膜细胞α-平滑肌肌动蛋白 (α- SMA)表达与细胞表型的关系。方法 :应用单克隆抗体 1- 2 2 - 3(Mo Ab1- 2 2 - 3)及 Habu蛇毒素分别诱导大鼠增殖性肾小球肾炎动物模型。用免疫组化技术并结合计算机图像分析 ,检测两个不同诱因引起模型的各个阶段的系膜细胞内α-平滑肌肌动蛋白 ,SMemb的表达程度 ,系膜细胞增殖和细胞外基质 (ECM)分泌的情况。结果 :在 Mo Ab1- 2 2 - 3模型 ,随着系膜细胞增殖程度的加重 ,ECM积聚增多 ,α- SMA的表达程度也增加 ,三者平行变化并具有高度相关性 ,而 Habu模型则系膜细胞增殖和 ECM增加的同时 α- SMA,SMemb表达不增加。结论 :α- SMA不是系膜细胞被激活并处于增殖 /分泌表型唯一标志
Objective: To investigate the relationship between the expression of α-smooth muscle actin (α-SMA) and the phenotype of mesangial cells in animal model of proliferative glomerulonephritis. Methods: The animal model of proliferative glomerulonephritis was induced by monoclonal antibody 1 - 2 2 - 3 (Mo Ab1-2 2 - 3) and Habu snake venom. Immunohistochemistry and computer image analysis were used to detect the expression of α-smooth muscle actin, SMemb, mesangial cell proliferation and extracellular matrix (ECM) secretion in mesangial cells in two phases induced by two different inducing factors Case. Results: With the increase of mesangial cell proliferation, the expression of ECM increased and the expression of α-SMA increased with the increase of mesangial cell proliferation in Mab Ab1-2-2-3 model. The Habu model was highly variable Membrane cell proliferation and ECM increase while α-SMA, SMemb expression did not increase. CONCLUSIONS: α-SMA is not the only marker of mesangial cells being activated and in a proliferative / secretory phenotype
