目的:探讨胃癌组织中EphA2与VEGF-C的表达及临床意义。方法:用免疫组化法检测82例胃癌组织及其癌旁组织中EphA2与VEGF-C蛋白的表达,以及淋巴管密度(LVD);用real-timePCR检测20例胃癌组织及其癌旁组织中EphA2与VEGF-Cm RNA的表达。分析EphA2与VEGF-C表达与患者临床病理特点及淋巴管生成的关系。结果:EphA2与VEGF-C在胃癌组织中的阳性表达率均明显高于癌旁正常组织(65.8%vs.42.6%;71.9%vs.39.0%,均P<0.05)。EphA2与VEGF-C的高表达与肿瘤浸润深度、淋巴结转移、TNM分期有关(均P<0.001)。EphA2蛋白与VEGF-C蛋白高表达的胃癌组织中LVD计数分别明显高于两者低表达胃癌组织(15.25±5.41vs.10.95±5.41,P=0.001;14.87±5.71vs.11.00±5.01,P=0.006)。胃癌组织中EphA2与VEGF-C蛋白以及m RNA表达均呈正相关(r=0.375,P=0.001;r=0.559,P=0.01)。结论:EphA2与VEGF-C在胃癌组织中均呈高表达,且可能共同促进淋巴管生成与胃癌淋巴结转移。
Objective: To investigate the expression of EphA2 and VEGF-C in gastric carcinoma and its clinical significance. Methods: The expressions of EphA2 and VEGF-C and lymphatic vessel density (LVD) were detected in 82 cases of gastric cancer tissues and paracancerous tissues by immunohistochemistry. In 20 gastric cancer tissues and their adjacent tissues EphA2 and VEGF-Cm RNA expression. The relationship between EphA2 and VEGF-C expression and clinicopathological features and lymphangiogenesis was analyzed. Results: The positive rates of EphA2 and VEGF-C in gastric cancer tissues were significantly higher than those in normal tissues (65.8% vs 42.6%; 71.9% vs.39.0%, both P <0.05). The overexpression of EphA2 and VEGF-C correlated with the depth of tumor invasion, lymph node metastasis and TNM staging (all P <0.001). The LVD counts in gastric cancer tissues with high expression of EphA2 protein and VEGF-C protein were significantly higher than those in both gastric cancer tissues (15.25 ± 5.41 vs.10.95 ± 5.41, P = 0.001; 14.87 ± 5.71 vs.11.00 ± 5.01, P = 0.006). There was a positive correlation between EphA2, VEGF-C protein and m RNA in gastric cancer tissues (r = 0.375, P = 0.001; r = 0.559, P = 0.01). Conclusion: Both EphA2 and VEGF-C are highly expressed in gastric carcinoma and may promote lymphangiogenesis and lymph node metastasis in gastric cancer.
