目的 利用杆状病毒 Ac MNPV载体在昆虫细胞中表达重组人胰岛素样生长因子 1( rh IGF- 1)。方法 含有胰岛素样生长因子 1( IGF- 1) c DNA的重组 Ac MNPV感染昆虫细胞株 Sf9及 Sf2 1后 ,采用酶联免疫吸附试验 ( EL ISA)检测被重组病毒感染的昆虫细胞表达产生的 rh IGF- 1,以野生型 Ac MNPV感染的昆虫细胞及空白细胞的细胞培养液、细胞裂解物作对照。结果 1重组 Ac MNPV感染的昆虫细胞大量表达有免疫活性的 rh IGF- 1,并可分泌至细胞培养液中 ;2受感染昆虫细胞 Sf2 1的培养上清中 rh IGF- 1含量较 Sf9高 ,而细胞裂解物则相反。结论 利用杆状病毒表达系统在昆虫细胞 Sf9和 Sf2 1中可大量表达 rh IGF- 1
Objective To express recombinant human insulin-like growth factor 1 (rh IGF-1) in insect cells using the baculovirus Ac MNPV vector. Methods Recombinant Ac MNPV harboring insulin-like growth factor 1 (IGF-1) c DNA was transfected into insect cell lines Sf9 and Sf2 1, and the enzyme-linked immunosorbent assay (ELISA) was used to detect the rh IGF-1 was used as a control for insect cells infected with wild-type Ac MNPV and cell culture medium and cell lysates of blank cells. Results 1 The recombinant Ac MNPV-infected insect cells expressed immunoreactive rh IGF-1 in large quantities and were secreted into the cell culture medium. 2 The content of rh IGF-1 in the supernatant of Sf21-infected insect cells was higher than that of Sf9, The cell lysate is the opposite. Conclusions Baculovirus expression system can express rh IGF-1 in insect cells Sf9 and Sf2 1
