目的 观察IL 2、4、6、7和IFN γ对γ射线诱导的人扁桃体淋巴细胞凋亡的调节作用。方法 取手术切除的扁桃体组织 ,经Ficoll分离得单个核细胞 ,采用 4Gy6 0 Coγ射线照射 ,用凝胶电泳显示细胞凋亡 ;体外用IL 2、IL 4、IL 6、IL 7及IFN γ与单核细胞共培养 2 4h后流式细胞仪测CD4+ 、CD8+ T细胞及CD19+ B细胞的凋亡率及表面bcl 2的表达。结果 照射后 4~ 2 4h有明显的梯形条带出现 ;CD8+ T细胞和B细胞的辐射敏感性较CD4+ T细胞高 ;IL 2、IL 4和IL 7对CD4+ 、CD8+ T细胞凋亡及IL 4对B细胞凋亡有明显的保护作用 ,同时可观察到bcl 2明显的高表达。IL 6和IFN γ无此作用。结论 细胞因子IL 2、IL 4与IL 7可促进放射病病人免疫功能恢复。
Objective To investigate the regulatory effect of IL 2,4,6,7 and IFN γ on γ-ray induced apoptosis in human tonsil lymphocytes. Methods Surgical excision of tonsil tissue was performed by using Ficoll to separate mononuclear cells. The cells were irradiated with 4Gy60Coγ ray and the cells were stained by gel electrophoresis. The cells were treated with IL 2, IL 4, IL 6, IL 7 and IFN γ in vitro Flow cytometry was used to measure the apoptosis rate and the expression of bcl-2 on CD4 +, CD8 + T cells and CD19 + B cells after co-cultured with nuclear cells for 24 hours. Results The obvious trapezoidal bands appeared 4 ~ 24 hours after irradiation. The radiosensitivity of CD8 + T cells and B cells was higher than that of CD4 + T cells. The apoptosis of CD4 + and CD8 + T cells by IL 2, IL 4 and IL 7, B cell apoptosis has a significant protective effect, while significant bcl 2 was observed high expression. IL 6 and IFN γ do not have this effect. Conclusion The cytokines IL 2, IL 4 and IL 7 can promote the recovery of immune function in patients with radiation sickness.
