目的 研究体外培养及成纤维细胞生长因子对骨髓基质细胞生长的影响。方法 抽取兔骨髓基质细胞体外培养 ,倒置显微镜动态观察 ,从原代培养 (P0 )中观察到细胞集落的形态有明显差异 ,提示骨髓基质细胞是一个由多种细胞组成的混合体。在培养液中加入碱性成纤维细胞生长因子 (bFGF ,5ng/ml)。 结果 第一代细胞(P1)的生长速度增至对照组的 1.5倍。而未经bFGF处理的细胞活跃增殖能力只能保持 4~ 5代 ,随后即发生退化 ;而经bFGF处理的细胞 ,传 10代以上仍能保持活跃的增殖能力。结论 bFGF能促进骨髓基质细胞增殖并长期保持其生物活性。
Objective To study the effects of cultured in vitro and fibroblast growth factor on the growth of bone marrow stromal cells. Methods Rabbit bone marrow stromal cells (BMSCs) were cultured in vitro and observed under inverted microscope. The morphological changes of colony were observed in primary culture (P0), suggesting that bone marrow stromal cells (BMSCs) are a mixture of many kinds of cells. Basic fibroblast growth factor (bFGF, 5 ng / ml) was added to the culture. Results The growth rate of the first generation cells (P1) was increased to 1.5 times that of the control group. However, the viability of cells without bFGF treatment maintained only 4 to 5 generations, followed by degeneration; and bFGF-treated cells, more than 10 generations can still maintain the active proliferation. Conclusion bFGF can promote the proliferation of bone marrow stromal cells and maintain its biological activity for a long time.
