目的:探讨八味柔肝颗粒对大鼠实验性肝纤维化的治疗作用。方法:以四氯化碳(CCl4)诱导大鼠肝纤维化模型。将大鼠随机分为八味柔肝颗粒治疗组(0.75,1.5,3 g·kg-1)、模型组、秋水仙咸碱组(0.5 mg·kg-1)和空白对照组。均连续ig 6周,观察各组肝组织中转移生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、金属蛋白酶组织抑制因子(TIMP-1)蛋白表达情况。结果:八味柔肝颗粒组TGF-β1,α-SMA,TIMP-1表达明显低于模型组(P<0.01)。结论:八味柔肝颗粒能抑制CCl4诱导实验性肝纤维化大鼠肝组织TGF-β1,α-SMA,TIMP-1的表达,可能是其抗肝纤维化的主要作用机制。
Objective: To explore the therapeutic effect of Bawei Rougan Granule on experimental hepatic fibrosis in rats. Methods: Carbon tetrachloride (CCl4) induced hepatic fibrosis in rats. The rats were randomly divided into eight treatment groups (0.75,1.5, 3 g · kg-1), model group, colchicine group (0.5 mg · kg-1) and blank control group. After 6 weeks, the expression of TGF-β1, α-SMA and TIMP-1 in each group were observed. Results: The expressions of TGF-β1, α-SMA and TIMP-1 in Bawei Rougan Granules group were significantly lower than those in model group (P <0.01). Conclusion: Bawei Rougan Granule can inhibit the expression of TGF-β1, α-SMA and TIMP-1 in CCl4-induced hepatic fibrosis rats, which may be the main mechanism of its anti-hepatic fibrosis.
