目的探讨乳清蛋白对胰岛素抵抗的改善作用。方法用高脂饲料制备胰岛素抵抗模型大鼠,将模型大鼠随机分为0%乳清蛋白(WP)组、5%WP组和15%WP组,每组10只。8周后测定大鼠空腹血糖(FBG)、空腹血浆胰岛素水平(FINS),进行葡萄糖耐量试验(OGTT)并计算葡萄糖曲线下面积(AUC),部分血浆用来测定总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)和丙二醛(MDA)等抗氧化指标。结果摄取乳清蛋白没有改变大鼠FBG和葡萄糖AUC。15%WP组大鼠血浆FINS水平和胰岛素抵抗指数(HOMA-IR)显著低于0%WP组(P<0.05)。15%WP组大鼠血浆T-AOC、SOD和GSH显著高于0%WP组(T-AOC:P<0.01;SOD、GSH:P<0.05),同时血浆MDA含量显著降低(P<0.05)。结论乳清蛋白有效改善了模型大鼠的胰岛素抵抗,这可能和增强机体抗氧化能力有关。
Objective To investigate the effect of whey protein on insulin resistance. Methods The model rats with insulin resistance were prepared with high-fat diet. The rats were randomly divided into 0% whey protein (WP) group, 5% WP group and 15% WP group, with 10 rats in each group. After 8 weeks, fasting plasma glucose (FBG) and fasting plasma insulin (FINS) were measured, glucose tolerance test (OGTT) and area under the curve of glucose (AUC) were calculated, and part of plasma was used to determine the total antioxidant capacity ), Superoxide dismutase (SOD), reduced glutathione (GSH) and malondialdehyde (MDA). Results Ingestion of whey protein did not alter rat FBG and glucose AUC. The levels of FINS and insulin resistance index (HOMA-IR) in 15% WP group were significantly lower than those in 0% WP group (P <0.05). The plasma levels of T-AOC, SOD and GSH in 15% WP group were significantly higher than those in 0% WP group (P <0.01); while the content of MDA in plasma was significantly lower (P <0.05) . Conclusions Whey protein can effectively improve the insulin resistance in model rats, which may be related to enhancing the antioxidant capacity of the body.
