目的:探讨化浊解毒活血方对急性心肌梗死(AMI)1周后心外膜梗死边缘区心肌细胞瞬间外向钾电流(Ito)的影响。方法:采用结扎冠状动脉左前降支的方法建立兔心肌梗死模型,30只兔随机分为3组:对照组,AMI组和化浊解毒活血方+AMI组。采用膜片钳技术研究化浊解毒活血方对左心室梗死边缘区心外膜(Epicardium,Epi)、心内膜(En-docardium,Endo)和中层(Midmycoardium,M)心室肌细胞的Ito密度的改变。结果:AMI组细胞的Ito电流密度明显降低,Epi与M、Endo之间差异较明显(P<0.01);化浊解毒活血方+AMI组使心外膜Ito电流密度进一步降低,三层细胞间无统计学意义(P>0.05)。结论:心肌梗死对Ito的跨壁异质性有明显影响;化浊解毒活血方减弱心肌梗死后Ito的跨壁异质性。
Objective: To investigate the effect of Huazhuo Jiedu Huoxue Recipe on myocardial outward transient potassium current (Ito) of epicardial infarction area 1 week after acute myocardial infarction (AMI). Methods: A rabbit model of myocardial infarction was established by ligation of left anterior descending coronary artery. Thirty rabbits were randomly divided into three groups: control group, AMI group and Huazhuo Jiedu Huoxue Fang + AMI group. Patch clamp technique was used to study the effects of Huazhuojieduhuoxue Recipe on the Ito density of epicardium (Epi), En-docardium (Endo) and midmycoardium (M) ventricular myocytes in the left ventricular infarct border region change. Results: The Ito current density of AMI group was significantly lower than that of E and M, Endo (P <0.01); the Huazhuo Jiedu Huoxue Recipe + AMI group further reduced the Ito current density of the epicardium, No statistical significance (P> 0.05). Conclusion: Myocardial infarction has a significant effect on the transmural heterogeneity of Ito. Huazhuo Jiedu Huoxue Fang can reduce the transmural heterogeneity of Ito after myocardial infarction.
