目的探讨T1DM患者血浆miR-126表达水平的变化及其临床意义。方法选取T1DM患者36例及健康对照(NGT)者34名,RT-PCR检测各组血浆miR-126,并分析其与年龄、BMI、高敏C反应蛋白(hsC-RP)、内皮型一氧化氮合酶(eNOS)、FPG、2hPG、HbA1c、FC-P、2hC-P、HDL-C、LDL-C、TG、TC的关系。结果与NGT组比较,T1DM组血浆miR-126表达下降[(1.65±0.61)vs(0.68±0.18),P<0.05]。相关分析显示,miR-126与HDL-C、FC-P、2hC-P及eNOS呈正相关(P<0.05或P<0.01),与LDL-C、TC、hsC-RP及年龄呈负相关(P<0.05或P<0.01);多元逐步回归分析显示,hsC-RP、FC-P、TC、年龄是miR-126的影响因素。结论 T1DM患者血浆miR-126呈低表达,可能与胰岛功能及内皮功能有关,hsC-RP及TC是miR-126的独立影响因素,miR-126可能参与了T1DM患者的炎症反应及脂质代谢异常,且miR-126水平随年龄增长而下降。
Objective To investigate the changes of plasma miR-126 level in patients with T1DM and its clinical significance. Methods Totally 36 patients with T1DM and 34 healthy controls (NGT) were enrolled in this study. Plasma miR-126 levels were measured by RT-PCR, and their relationships with age, BMI, hsC-RP and endothelial nitric oxide (ENOS), FPG, 2hPG, HbA1c, FC-P, 2hC-P, HDL-C, LDL-C, TG and TC. Results Compared with NGT group, plasma miR-126 level in T1DM group was significantly lower than that in NGT group [(1.65 ± 0.61) vs (0.68 ± 0.18), P <0.05]. Correlation analysis showed that miR-126 was positively correlated with HDL-C, FC-P, 2hC-P and eNOS (P <0.05 or P <0.01) and negatively correlated with LDL-C, TC, <0.05 or P <0.01). Multiple stepwise regression analysis showed that hsC-RP, FC-P, TC and age were the influencing factors of miR-126. Conclusion The low expression of plasma miR-126 in T1DM patients may be related to islet function and endothelial function. HsC-RP and TC are independent risk factors for miR-126. MiR-126 may be involved in inflammatory reaction and lipid metabolism in T1DM patients , And the level of miR-126 decreased with age.
