目的:建立糖尿病性脑梗死大鼠模型,观察对比高级糖基化终产物受体(RAGE)在模型大鼠中的表达及活络育阴方对其的影响。方法:选SD大鼠以高脂高糖饮食,腹腔注射链脲佐菌素(STZ)诱导建立糖尿病模型,在此基础上再用线栓法制作大脑中动脉栓塞(MCAO)模型;利用形态学等方法,观察不同剂量和不同时间活络育阴方对糖尿病性脑梗死的脑保护作用;用ABC免疫组化方法观察活络育阴方对糖尿病性脑梗死模型大鼠脑损伤后RAGE表达的影响。结果:活络育阴方治疗糖尿病性脑梗死大鼠,在不同剂量和不同时间内大鼠脑组织RAGE表达均明显降低。结论:活络育阴方可能通过下调RAGE表达,对糖尿病性脑梗死大鼠的神经损伤起到保护作用。
OBJECTIVE: To establish a rat model of diabetic cerebral infarction and to observe the expression of advanced glycation endproduct receptor (RAGE) in model rats and the effect of active collaterals and yin-yuyin on it. METHODS: SD rats were induced by intraperitoneal injection of streptozotocin (STZ) on a high-fat and high-sugar diet to establish a diabetes model. Then, a middle cerebral artery occlusion (MCAO) model was made by using a thread embolization method. Morphology was used. Methods: To observe the neuroprotective effect of Huoluo Yuyin Decoction at different doses and at different times on diabetic cerebral infarction; observe the effect of Huoluo Yuyin Decoction on the expression of RAGE after cerebral injury in diabetic rat model with ABC immunohistochemistry. RESULTS: After treated with Huoluo Yuyin Decoction, the expression of RAGE in brain tissue of rats with different doses and at different times was significantly decreased. Conclusion: Huoluoyuyin Decoction may protect neurological injury in rats with diabetic cerebral infarction by down-regulating RAGE expression.
