用细胞核仁组成区相关蛋白染色与图像分析技术,研究γ干扰素(IFNγ)对正常和不同因子刺激的NIH/3T3成纤维细胞相关嗜银蛋白(AgNORs)的影响。发现(1)不同浓度的IFNγ对正常成纤维细胞的生长显示出明显的抑制作用,其中以250×103u/L浓度IFNγ的抑制作用较强(P<001);(2)IFNγ对肝素、5羟色胺和IL1刺激的成纤维细胞增殖的影响表明,250×103u/L浓度的IFNγ对肝素和5羟色胺刺激的成纤维细胞显示出明显的抑制作用,其AgNORs三个参数的均值相当于对照值的54%和51%,而250和500×103u/L浓度的IFNγ能明显抑制IL1刺激的成纤维细胞生长,其抑制率分别为对照值的61%和55%(P<001);(3)比较IFNγ对正常和刺激生长的成纤维细胞的作用,发现IFNγ能不同程度地抑制成纤维细胞的生长,其中对5羟色胺刺激的成纤维细胞抑制作用最强,其次为正常和IL1刺激的成纤维细胞,而对肝素刺激的成纤维细胞增生的抑制作用较弱,其抑制率分别为对照值的63%,66%,73%和85%。以上结果为IFNγ治疗各种因素,特别是辐射引起的器官纤维化研究提供了?
The effects of interferon-gamma (IFN-γ) on NIH / 3T3 fibroblast-associated argyrophilic proteins (AgNORs) stimulated by normal and different factors were studied by staining the nucleolar organizer region-related proteins and imaging analysis.
