目的观察慢性砷中毒对小鼠海马齿状回超微结构的改变及小鼠学习记忆的影响。方法选取健康成年昆明小鼠80只,雌雄各半,分为对照组、慢性砷中毒低、中、高剂量组,每组20只,各染砷组分别以蒸馏水、1/5LD50、1/10LD50、1/40LD50As2O3灌胃,连续3个月,根据其体重变化随时调整用药剂量。灌胃结束后,采用Y-电迷宫检测各组小鼠学习记忆行为,透射电镜观察齿状回细胞超微结构的变化。结果高剂量砷中毒组学习、记忆的Y-迷宫测试次数明显高于对照组(P<0.05),电镜下观察中毒组齿状回神经元,可见部分神经细胞膜皱缩,胞体减小,核染色质电子密度增高。结论慢性砷中毒明显损害成年小鼠学习记忆的能力,可能与海马齿状回神经元受损有关。
Objective To observe the changes of ultrastructure of dentate gyrus and the learning and memory of mice after chronic arsenism. Methods Totally 80 adult Kunming mice, male and female, were divided into control group, chronic arsenism low, medium and high dose group, 20 rats in each group. The arsenic groups were treated with distilled water, 1 / 5LD50 and 1 / 10LD50 , 1 / 40LD50As2O3 gavage for 3 consecutive months, according to their weight changes at any time to adjust the dosage. At the end of gavage, the learning and memory behaviors of mice in each group were detected by Y-electric maze and the ultrastructural changes of dentate gyrus cells were observed by transmission electron microscope. Results The numbers of learning and memory Y-maze test in high dose arsenic poisoning group were significantly higher than those in control group (P <0.05). The neurons in dentate gyrus of poisoning group were observed under electron microscope. Elevated electron density. Conclusion Chronic arsenic poisoning significantly impaired the learning and memory abilities of adult mice, which may be related to the damage of dentate gyrus neurons in the hippocampus.
