目的 建立大鼠中脑多巴胺神经元的培养体系 ,判断百草枯 (Paraquat)和氧桥氯甲桥萘 (Dieldrin)能否对培养的大鼠中脑多巴胺神经元产生选择性损害。方法 将 0 .0 0 1~ 1 0 0 μmol/L浓度的Paraquat、Dieldrin加入到原代培养的大鼠中脑多巴胺神经元中 ,计数存活的多巴胺神经元和非多巴胺神经元。结果 Paraquat对大鼠中脑多巴胺神经元无选择性损害。 1 μmol/LDieldrin能选择性损害大鼠中脑多巴胺神经元。 结论 Dieldrin可能是致帕金森病 (PD)的神经毒物 ,它与PD病因之间的联系值得进一步研究
OBJECTIVE: To establish a culture system of dopamine neurons in the rat midbrain to determine whether paraquat and Dieldrin selectively damage dopaminergic neurons in cultured rat brain. Methods Paraquat and Dieldrin were added into the cultured rat mesencephalic dopaminergic neurons at a concentration of 0.010-1.00 μmol / L, and survived dopamine neurons and non-dopamine neurons were counted. Results Paraquat has no selective damage to rat midbrain dopamine neurons. 1 μmol / LDieldrin can selectively damage dopaminergic neurons in the rat midbrain. Conclusions Dieldrin may be a neurotoxic agent of Parkinson’s disease (PD), and its association with the etiopathogenis of PD deserves further study
