目的 :研究高温对原代培养神经细胞存活的影响以及bFGF和NGF对高温处理后神经细胞存活的保护作用。方法 :应用神经细胞原代培养、MTT、NSE ELISA测定技术。结果 :①高温处理后神经细胞活性明显降低 ;②外源性bFGF、NGF均不同程度地增加了神经细胞活性 ,其抗体均不同程度地使神经细胞活性降低 ,但二者联合应用神经细胞活性增加不明显 ;③NSE ELISA检测结果与MTT检测结果一致 ,但特异性和敏感性更高。结论 :外源性bFGF和NGF对高温处理后神经细胞存活具有保护作用。
OBJECTIVE: To investigate the effects of high temperature on the survival of primary cultured neurons and the protective effect of bFGF and NGF on the survival of neurons after high temperature treatment. Methods: Primary culture of neurons, MTT and NSE ELISA were used. Results: ①Nerve cell activity was significantly decreased after hyperthermia treatment; ②Exogenous bFGF and NGF increased neuronal activity in varying degrees, and their antibodies reduced neuronal activity to varying degrees, but the combination of the two increased neuronal activity Not obvious; ③NSE ELISA test results and MTT test results, but specificity and sensitivity higher. Conclusion: Exogenous bFGF and NGF have a protective effect on the survival of nerve cells after high temperature treatment.
