应用免疫组织化学和原位杂交方法检测人消化道癌患者胃肠道粘膜中的MUC2 和MUC3 基因蛋白产物和mRMA 的表达,以提示MUC2 和MUC3 基因在人消化道癌患者胃肠道粘膜中的表达规律。结果发现,胃粘膜中无MUC2 和MUC3 基因产物的表达,MUC2 核粘蛋白及mRNA 主要存在十二指肠和结肠杯状细胞周及核上,柱状细胞中无表达。MUC3 核粘蛋白及其mRNA 主要存在十二指肠和结肠的杯状细胞和柱状细胞胞浆内,而杯状细胞的粘液滴内无MUC2、MUC3;基因产物的阳性反应信号。提示,MUC2 和MUC3 基因主要在肠道粘膜中表达,而胃粘膜中不表达,是合成肠道粘液的主要粘蛋白基因。
Immunohistochemistry and in situ hybridization were used to detect the expression of MUC2 and MUC3 gene protein products and mRMA in the gastrointestinal mucosa of patients with human digestive tract cancer, in order to suggest that the MUC2 and MUC3 genes are in the gastrointestinal mucosa of patients with human gastrointestinal cancer. Expression rules. The results showed that there was no expression of MUC2 and MUC3 gene products in the gastric mucosa, and MUC2 nuclear mucin and mRNA were mainly present in the duodenum and colon goblet cells and on the nucleus, and no expression was observed in the columnar cells. MUC3 nuclear mucin and its mRNA were mainly present in the cytoplasm of goblet and columnar cells of the duodenum and colon, but there was no MUC2, MUC3 in the goblet cells of the goblet cells; positive reaction signals of the gene products. It is suggested that the MUC2 and MUC3 genes are mainly expressed in the intestinal mucosa but not in the gastric mucosa and are the major mucin genes for the synthesis of intestinal mucus.
