为探讨牛磺酸对肝脏保护作用的机理,本实验观察了牛磺酸对四氯化碳(CCl_4)肝坏死大鼠血浆内皮素(Endothelin.ET)的影响.1 材料和方法取♂Wistar大鼠(体重180~220g).实验前24h禁食.用CCl_4原液以2.5ml·kg~(-1)剂量ig.复制肝坏死模型.保护组除给予CCl_4外,预先2d给予牛磺酸250mg·kg~(-1)·d~(-1)背部sc;对照组在同等条件下sc10ml·kg~(-1)生理盐水.各组动物ig后24h取血测定血浆ET含量、谷丙转氨酶(ALT)和乳酸脱氢酶(LDH)活性,取肝脏测定肝组织丙二醛(MDA)含量、组织钙和线粒体钙含量.
In order to explore the mechanism of taurine on liver protection, we observed the effect of taurine on endothelin (ET) in liver of rats with carbon tetrachloride (CCl_4) hepatic injury.1 Materials and Methods Rats (weight: 180-220g) were fasted for 24 hours before the experiment, and the model of hepatic necrosis was reproduced with ig of 2.5ml · kg ~ (-1) with CCl_4 solution.After 2 weeks’ administration of CCl_4, (-1) · d ~ (-1) dorsal sc and control group under the same conditions sc10ml · kg ~ (-1) saline.The blood samples were collected at 24h after ig to determine the content of ET, the content of alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) activity were measured. The content of malondialdehyde (MDA), tissue calcium and mitochondrial calcium in the liver were measured.
