采用在骨片上培养日本大耳白兔破骨细胞的方法,研究了不同浓度淫羊藿苷(icariin)对破骨细胞骨吸收功能的影响.为了定量评价破骨细胞的骨吸收功能和探讨淫羊藿苷的作用机制,采用显微摄影结合计算机图像分析技术测定了骨吸收造成的陷窝表面积,并检测了细胞肌动蛋白环(actin-ring)、细胞内游离钙离子的浓度[Ca2+]i和超氧阴离子自由基(~·O2-)的变化.结果显示,淫羊藿苷降低了破骨细胞内的钙离子浓度,这可能是引起肌动蛋白环回缩和细胞内超氧阴离子自由基减少的原因,进而导致吸收陷窝面积减小,抑制了破骨细胞的骨吸收.
The effect of different concentrations of icariin on the bone resorption of osteoclasts was studied by culturing osteoclasts from Japanese big-eared rabbits on bone chips.
