目的:观察熊果酸(Ursolic acid,UA)是否通过激活卵巢癌SKOV3细胞的线粒体凋亡通路诱导细胞凋亡。方法:MTT法检测细胞增殖抑制,采用流式细胞术检测细胞凋亡和线粒体膜电位,免疫印迹法检测细胞色素C的表达,分光光度法检测Caspase-9、Caspase-3的活性。结果:熊果酸可以抑制SKOV3细胞增殖,40μmol/L熊果酸,作用于SKOV3细胞48 h,增殖抑制率可以达到76.85±3.1%;UA可以诱导细胞发生凋亡;熊果酸可以降低SKOV3细胞的线粒体膜电位,促进细胞色素C的表达,增强Caspase-9、Caspase-3的活性。结论:熊果酸可以促进SKOV3细胞的凋亡,可能的作用机制之一是可以激活SKOV3细胞的线粒体凋亡通路。
AIM: To investigate whether ursolic acid (UA) induces apoptosis in ovarian cancer cell line SKOV3 by activating the mitochondrial apoptotic pathway. Methods: MTT assay was used to detect the cell proliferation. Cell apoptosis and mitochondrial membrane potential were measured by flow cytometry. The expression of cytochrome C was detected by Western blotting. The activity of Caspase-9 and Caspase-3 was detected by spectrophotometry. Results: Ursolic acid could inhibit the proliferation of SKOV3 cells and inhibit the proliferation of SKOV3 cells by 40μmol / L ursolic acid for 48 h. The inhibition rate of ursolic acid could be 76.85 ± 3.1%. UA can induce cell apoptosis. Ursolic acid can decrease SKOV3 cells Mitochondrial membrane potential, promote the expression of cytochrome C and enhance the activity of Caspase-9, Caspase-3. Conclusion: UA can promote the apoptosis of SKOV3 cells. One possible mechanism is that it can activate the mitochondrial apoptotic pathway of SKOV3 cells.
