目的探讨模拟微重力对人脐静脉内皮细胞(HUVEC)生成血管生成素(ANG)的影响。方法培养 HUVEC,分为模拟微重力组、地面静止对照组及剪切力对照组,在培养第24h、48h、72h 和96h 分别收集培养上清液,用 ELISA 方法检测 ANG 的浓度变化;固定一部分细胞,应用免疫细胞化学方法检测 ANG 在 HUVEC 中的表达情况。结果无论是应用 ELISA 方法检测上清液中的 ANG,还是应用免疫细胞化学的方法检测 ANG 在细胞内的表达,均发现与地面静止及剪切力对照组相比,模拟微重力组 HUVEC 的 ANG 生成增加(P<0.05)。结论应用回转器模拟微重力能促进体外培养的HUVEC 生成 ANG。
Objective To investigate the effects of simulated microgravity on angiogenin (ANG) production in human umbilical vein endothelial cells (HUVECs). Methods HUVECs were cultured and divided into simulated microgravity group, ground static control group and shear stress control group. Culture supernatants were harvested at 24h, 48h, 72h and 96h after culture respectively. The concentration of ANG was detected by ELISA. The expression of ANG in HUVECs was detected by immunocytochemistry. Results Whether using the ELISA method to detect ANG in the supernatant or detecting the expression of ANG in the cells by immunocytochemistry, we found that compared with the gestation and shear force control group, the simulated microgravity HUVEC ANG Generation increased (P <0.05). Conclusion The application of gyrator to simulate microgravity can promote the generation of ANG in cultured HUVECs.
