利用AB-8型大孔吸附树脂柱色谱、ODS C_(18)柱色谱、Sephadex LH-20柱色谱和制备型高效液相色谱,从广东紫珠Callicarpa kwangtungensis Chun地上部位60%乙醇提取物中分离得到4个黄酮二葡萄糖醛酸苷类化合物,通过理化性质和波谱分析鉴定结构,确定分别为金合欢素-7-O-[?-D-吡喃葡萄糖醛酸基(1→2)-O-?-D-吡喃葡萄糖醛酸苷](1),金圣草素-7-O-[?-D-吡喃葡萄糖醛酸基(1→2)-O-?-D-吡喃葡萄糖醛酸苷](2),芹菜素-7-O-[?-D-吡喃葡萄糖醛酸基(1→2)-O-?-D-吡喃葡萄糖醛酸苷](3)和木犀草素-7-O-[?-D-吡喃葡萄糖醛酸基(1→2)-O-?-D-吡喃葡萄糖醛酸苷](4)。化合物1~4均为首次从紫珠属植物中分离得到。采用基于荧光强度检测牛源环氧合酶(COX)-1和人源COX-2的方法评价化合物的抗炎活性,结果化合物2~4均表现出一定的COX抑制活性,其中化合物4活性较强。
The crude extracts were separated from 60% ethanol extract of aerial parts of Callicarpa kwangtungensis Chun by using AB-8 macroporous resin column chromatography, ODS C 18 column chromatography, Sephadex LH-20 column chromatography and preparative high performance liquid chromatography Four flavone diglucuronide compounds were obtained. Their structures were identified by physico-chemical properties and spectral analysis. Their structures were identified as acacetin-7-O - [? - D-glucopyranosyl -? - D-glucopyranoside] (1), rhamnoside-7-O - [? - D-glucopyranosyl (1 → 2) -O-? - D-pyran Glucuronide] (2), apigenin-7-O - [? - D-glucopyranosyl (1 → 2) -O-? -D- glucopyranoside] (3) and Luteolin-7-O - [? - D-glucopyranosyl (1 → 2) -O -? - D-glucopyranoside] (4). Compounds 1 to 4 were isolated from the genus Umbelliferus for the first time. The anti-inflammatory activity of the compounds was evaluated based on the fluorescence intensity detection of bovine COX-1 and human COX-2. As a result, compounds 2 to 4 showed some COX inhibitory activity, of which compound 4 exhibited strong activity.
