用硫酸铵分级沉淀和制备性电泳 ,从赤子爱胜蚓 (Eiseniafoelida)粗制品中纯化出 5种组分 (Ⅰ、Ⅶ、Ⅷ、Ⅸ、Ⅹ ) ,在PAGE中均呈单一蛋白带。SPS_PAGE测得分子量分别为32 0 0 0、2 6 5 0 0、2 6 0 0 0、2 45 0 0和 2 1 5 0 0。等电点为 3.80~ 4.2 5不等 ,最大吸收峰均在 2 76nm。 5种组分的最适pH在 8.5左右 ,最适温度为 6 0± 2℃。在 pH6 .0~ 1 0 .0和 6 0℃以下有较高的稳定性 ,组分Ⅰ在 70℃、1 0min活性不变 ,对某些变性剂表现很强耐受性。组分Ⅰ的活性不能被贵州小慈菇蛋白酶抑制剂抑制
Using ammonium sulfate fractionation precipitation and preparative electrophoresis, five components (I, VII, VIII, IX, X) were purified from the crude product of Eisenia foelida, and each protein was a single protein band in PAGE. The molecular weights measured by SPS_PAGE were 32 0 0 0, 2 6 500, 2 600, 2 4500, and 2 1 500, respectively. The isoelectric point ranged from 3.80 to 4.2 5, and the maximum absorption peaks were all at 2 76 nm. The optimum pH of the five components is around 8.5, and the optimum temperature is 60±2°C. At pH 6.0 - 10.0 and below 60°C, there is high stability. Component I has no change in activity at 70°C and 10 min, and it is very resistant to certain denaturants. Component I activity cannot be inhibited by S. gigas protease inhibitors
