目的研究肝心宁对肝纤维化大鼠肝组织α-平滑肌肌动蛋白(α-SMA)、基质金属蛋白酶-13(MMP-13)及金属蛋白酶组织抑制剂-1(TIMP-1)蛋白表达的影响。方法将68只雄性SD大鼠随机分为正常组、模型组、秋水仙碱组、肝心宁高剂量组、肝心宁低剂量组。采用四氯化碳皮下注射并饲以高脂低蛋白饲料和30%酒精诱导大鼠肝纤维化模型。各组分别处理,6周末用免疫组织化学染色法检测肝组织中α-SMA,MMP-13及TIMP-1蛋白表达情况。结果肝心宁高剂量组、肝心宁低剂量组和秋水仙碱组与模型组相比,α-SMA及TIMP-1均明显降低,P<0.05;肝心宁高剂量组与肝心宁低剂量组和秋水仙碱组相比,α-SMA及TIMP-1均明显降低,P<0.05;肝心宁高剂量组、肝心宁低剂量组和秋水仙碱组与模型组相比,MMP-13显著升高,P<0.05;肝心宁高剂量组与肝心宁低剂量组和秋水仙碱组相比,MMP-13显著升高,P<0.05。结论肝心宁抑制大鼠肝脏纤维组织的形成可能与其降低α-SMA,TIMP-1蛋白表达并增强MMP-13蛋白表达有关。
Objective To investigate the effect of Ganxinning on the expression of α-smooth muscle actin (α-SMA), matrix metalloproteinase-13 (MMP-13) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in rat liver fibrosis. influences. Methods Sixty-eight male SD rats were randomly divided into normal group, model group, colchicine group, high-dose Ganxinning group and low-dose Ganxinning group. The hepatic fibrosis model was induced by subcutaneous injection of carbon tetrachloride and fed with high-fat low-protein diet and 30% alcohol. Each group was treated separately. The expression of α-SMA, MMP-13 and TIMP-1 protein in liver tissue was detected by immunohistochemical staining at the end of 6 weeks. Results Compared with model group, high-dose Ganxinning group, low-dose Ganxinning group, and colchicine group had significantly decreased α-SMA and TIMP-1, P<0.05; high-dose Ganxinning group and low-dose Ganxinning group Compared with the colchicine group, α-SMA and TIMP-1 were significantly lower, P<0.05; the high-dose Ganxinning group, Ganxinning low-dose group, and colchicine group were significantly higher than those of the model group. ,P<0.05; Compared with the low-dose Ganxinning group and the colchicine group, MMP-13 significantly increased in the high-dose Ganxinning group, P<0.05. Conclusion Ganxining inhibits the formation of hepatic fibrous tissue in rats may be related to the decrease of α-SMA, TIMP-1 protein expression and enhancement of MMP-13 protein expression.
