目的:观察苦参碱对宫颈癌HeLa细胞中JAK-STAT信号通路的负性调控作用。方法:以不加药细胞为对照组,实验组加入不同浓度苦参碱(0.5、1.0、2.0 mg/m L)作用24 h,Western blot法检测SHP2、SOCS3、PIAS1蛋白表达、Real-Time PCR检测SHP2、SOCS3、PIAS1的mRNA表达。结果:与对照组比较,各给药组细胞SHP2、SOCS3蛋白及mRNA表达显著上调(P<0.05),其作用呈浓度依赖性;各组PIAS1蛋白及mRNA表达差异均无统计学意义(P>0.05)。结论:苦参碱通过上调HeLa细胞中SHP2、SOCS3蛋白及mRNA水平,负性调节JAK-STAT信号通路活性,抑制宫颈癌HeLa细胞增殖。
Objective: To observe the negative regulation effect of matrine on JAK-STAT signal pathway in cervical cancer HeLa cells. Methods: The untreated cells were used as the control group. Matrine (0.5,1.0,2.0 mg / ml) was added to the experimental group for 24 h. The protein expression of SHP2, SOCS3 and PIAS1 were detected by Western blot. Real-time PCR The mRNA expressions of SHP2, SOCS3 and PIAS1 were detected. Results: Compared with the control group, the protein and mRNA expressions of SHP2 and SOCS3 in each group were significantly increased (P <0.05), and the effect was concentration-dependent. There was no significant difference in the expression of PIAS1 protein and mRNA in each group (P> 0.05). Conclusion: Matrine inhibits the proliferation of cervical cancer HeLa cells by up-regulating SHP2, SOCS3 protein and mRNA levels in HeLa cells, negatively regulating the activity of JAK-STAT signaling pathway.
