目的:探讨雄激素对前列腺癌细胞和组织中miR-204表达的影响。方法:采用荧光定量PCR方法检测前列腺癌细胞株(LNCaP、22RV1)和正常前列腺间质细胞(WPMY-1)在雄激素类似物R1881刺激下以及去势后不同时间的小鼠前列腺组织中miR-204表达量的变化。结果:在前列腺癌细胞LNCaP、22RV1中hsa-miR-204的表达量在雄激素刺激后下降了近2倍,而在去势小鼠的前列腺组织中,mmu-miR-204表达量增加了1.8倍。结论:miR-204受雄激素的负调控。
Objective: To investigate the effect of androgens on the expression of miR-204 in prostate cancer cells and tissues. Methods: Fluorescence quantitative PCR was used to detect the expression of miR-1 in prostate cancer cells (LNCaP, 22RV1) and normal prostatic stromal cells (WPMY-1) under the stimulation of androgen analogue R1881 and at different time points after castration. 204 expression level changes. Results: The expression of hsa-miR-204 in prostate cancer cells LNCaP and 22RV1 decreased by nearly 2-fold after androgen stimulation, while in the prostate tissue of ovariectomized mice, the expression of mmu-miR-204 increased by 1.8 Times Conclusion: miR-204 is negatively regulated by androgens.
