组分中药KX组合物及单味组分对实验性自身免疫性心肌炎模型的影响

目的:研究组分中药KX组合物及其等剂量的单味组分对实验性自身免疫性心肌炎(EAM)模型的影响。方法:制备EAM模型,于首次免疫后2、8周,分别测定血液中IL-2、IL-17、TNF-α、IFN-γ的含量,取心脏做病理检查。结果:注射抗原2周后,模型大鼠血液中IL-2、IL-17、TNF-α、IFN-γ明显升高(P<0.05,P<0.01)。8周时IFN-γ持续升高(P<0.05),其余因子水平恢复正常。心肌由2周的细胞变性、炎细胞浸润逐渐演变为组织纤维化、坏死。苦参总碱给药2周时降低血液中IL-17、TNF-α、IFN-γ(P<0.01);8周时IL-17、TNF-α持续降低(P<0.01,P<0.05)。西洋参总皂苷2周时降低了IL-2、IL-17、TNF-α、IFN-γ,8周时IL-2、IL-17、IFN-γ仍降低。KX组合物2周时降低了IL-2、IL-17、TNF-α、IFN-γ(P<0.01,P<0.05),并持续到8周(P<0.01)。病理结果表明,各给药组心肌损伤程度均轻于模型组。结论:KX组合物及其单味组分能不同程度的明显抑制EAM模型Th1、Th17细胞亚群激活的相关细胞因子的释放,减轻心肌损伤;组合物的作用效果体现了中药的配伍优势。 Objective: To study the effect of component KX composition and its single dose of single component on experimental autoimmune myocarditis (EAM) model. Methods: The EAM model was prepared. The levels of IL-2, IL-17, TNF-α and IFN-γ in the blood were measured at 2 and 8 weeks after the first immunization. The heart was taken for pathological examination. Results: The levels of IL-2, IL-17, TNF-α and IFN-γ in the blood of model rats were significantly increased after 2 weeks of antigen injection (P <0.05, P <0.01). After 8 weeks IFN-γ continued to increase (P <0.05), the rest of the factors returned to normal levels. Myocardial cells degeneration by 2 weeks, inflammatory cell infiltration gradually evolved into tissue fibrosis, necrosis. The levels of IL-17, TNF-α and IFN-γ in the blood were decreased 2 weeks after the administration of matrine and total alkaloids (P <0.01) . The ginsenosides reduced IL-2, IL-17, TNF-α and IFN-γ at 2 weeks, and still decreased IL-2, IL-17 and IFN-γ at 8 weeks. KX compositions decreased IL-2, IL-17, TNF-α, IFN-γ by 2 weeks (P <0.01, P <0.05) for up to 8 weeks (P <0.01). The pathological results showed that the degree of myocardial injury in each administration group was lighter than that in the model group. CONCLUSION: KX and its single component can significantly inhibit the release of cytokines activated by Th1 and Th17 cell subsets in EAM model to a certain extent, and relieve myocardial injury. The effect of the composition reflects the compatibility advantage of traditional Chinese medicine.
