用重氮偶合试剂进行分光法测定普鲁卡因胺血清浓度的方法及其稳定性考察,结果日内和日间误差较小(CV<4%),操作方法较简便,适用于临床常规检测。通过对家兔药动学实验研究,其主要药动学参数α=7.04±0.93h~(-1),B:0.56±0.09h~(-1),K_(10)=1.91±0.18h~(-1),K_(12)=2.06±0.37h~(-1),V_c=2.42±0.32L/kg,V_d=8.76±2.36,C1=4.62±0.75,符合二房室模型,药动学方程为C=36.23e~(-7.04t)+11.14e~(-0.559t),实验表明其理论曲线与实验曲线拟合较好。
Spectrophotometric determination of procainamide serum with diazo coupling reagent method and its stability study, the results of intra-day and inter-day error is small (CV <4%), the operation method is simple and suitable for routine clinical testing. The main pharmacokinetic parameters were as follows: α = 7.04 ± 0.93h ~ (-1), B: 0.56 ± 0.09h ~ (-1), K_ (10) = 1.91 ± 0.18h ~ (-1) K_ (12) = 2.06 ± 0.37h ~ (-1), V_c = 2.42 ± 0.32L / kg, V_d = 8.76 ± 2.36, C1 = 4.62 ± 0.75, C = 36.23e ~ (-7.04t) +11.14e ~ (-0.559t). The experimental results show that the theoretical and experimental curves fit well.
