目的 :用药理学方法研究古方补阳还五汤颗粒剂的药效学及其急性毒性 ,为治疗缺血性中风提供理论依据。方法 :补阳还五汤颗粒剂对实验性大鼠脑缺血的作用 ,对大鼠心电、血压、颈动脉血流量的影响 ,对凝血时间及血栓的影响。结果 :该颗粒剂对脑缺血大鼠脑指数显著降低 (P <0 .0 5) ,使颈总动脉血流量增加 (P <0 .0 1 ) ,降低脑血管阻力 (P <0 .0 1 ) ,延长凝血时间及抑制血栓的形成 (P <0 .0 1 ) ;急性毒性试验表明 ,对小鼠ig最大耐受量大于 2 7 5g·kg- 1 ,iv最大耐受量大于 3g·kg- 1 。结论 :补阳还五汤颗粒剂可增加颈总动脉血流量 ,降低脑血管阻力 ,延长凝血时间及抑制血栓的形成 ,对治疗脑缺血诱发的中风有一定意义 ,是一安全有效的古方制剂
Objective: To study the pharmacodynamics and acute toxicity of Gufang Buyang Huanwu Decoction Granules with pharmacological methods, and provide a theoretical basis for the treatment of ischemic stroke. Methods: The effect of Buyang Huanwu Decoction Granule on experimental cerebral ischemia in rats, the influence of electrocardiogram, blood pressure, carotid blood flow in rats, and the clotting time and thrombus. Results: The granules significantly reduced the cerebral index of cerebral ischemia in rats (P < 0.05), increased the blood flow of common carotid arteries (P <0.01), and reduced cerebral vascular resistance (P <0. 1) prolonged clotting time and inhibited the formation of thrombus (P < 0.01); acute toxicity test showed that the maximal tolerance of mice is greater than 275g·kg-1, and the iv maximum tolerance is greater than 3g. Kg-1. Conclusion: Buyang Huanwu decoction granules can increase blood flow of common carotid artery, reduce cerebral vascular resistance, prolong clotting time and inhibit the formation of thrombus. It has certain significance for the treatment of stroke induced by cerebral ischemia. It is a safe and effective ancient prescription.
