目的采用HPLC法测定牡丹皮中三没食子酰葡萄糖和五没食子酰葡萄糖的含量。方法采用Agilent TC-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%甲酸,梯度洗脱,流速0.8 m L·min~(-1),检测波长274 nm,柱温25℃,进样量10μL。结果三没食子酰葡萄糖0.068~0.680μg(r=0.9994)、五没食子酰葡萄糖0.604~6.040μg(r=0.9997)与峰面积呈良好的线性关系,二者的平均加样回收率分别为99.31%、99.11%,RSD分别为1.48%、1.22%(n=9)。结论所用方法简便、准确、稳定、灵敏度高,可用于牡丹皮药材的质量控制。
OBJECTIVE To determine the content of triamino glucosamine and decalloyl glucose in Cortex Moutan by HPLC. Methods The mobile phase consisted of acetonitrile-0.1% formic acid with gradient elution at a flow rate of 0.8 mL · L -1 on an Agilent TC-C 18 column (250 mm × 4.6 mm, 5 μm) with a detection wavelength of 274 nm , Column temperature 25 ℃, injection volume 10μL. Results The results showed that there was a good linear relationship between the concentration of triagloylglucose 0.068-0.680μg (r = 0.9994) and pentadecanoylglucose 0.604-6.040μg (r = 0.9997) and the peak area. The average recoveries of the two compounds were 99.31% 99.11% with RSDs of 1.48% and 1.22% respectively (n = 9). Conclusion The method is simple, accurate, stable and sensitive. It can be used for the quality control of Cortex Moutan.
