目的同时测定阴洗液中咖啡酸和蛇床子素的含量。方法采用梯度洗脱HPLC法测定咖啡酸和蛇床子素的含量。色谱柱:C_(18)(Kromasil,250 mm×4.6 mm,5μm);流动相:A:0.01 mol/L磷酸盐缓冲液(pH4.0),B:乙腈;流速:1.0 m L/min;检测波长:322 nm;柱温:35℃。结果咖啡酸和蛇床子素与其他成分分离完全,峰形良好。咖啡酸在0.02~0.26μg范围内线性关系良好(r=0.999 6)。平均加样回收率为97.8%,RSD为1.7%(n=9);蛇床子素在0.04~0.39μg范围内线性关系良好(r=0.999 9)。平均加样回收率为98.0%,RSD为0.9%(n=9)。结论所建立的方法简便、准确、重现性好,可作为该制剂中2种成分的质量控制。
Objective To determine the content of caffeic acid and osthole in the lotion. Methods The content of caffeic acid and osthole were determined by gradient elution HPLC. Column: C_ (18) (Kromasil, 250 mm × 4.6 mm, 5 μm); mobile phase: A 0.01 mol / L phosphate buffer pH 4.0 B: acetonitrile flow rate 1.0 m L / min; Detection wavelength: 322 nm; column temperature: 35 ℃. Results caffeic acid and osthole were completely separated from other components, with good peak shape. Caffeic acid showed good linearity in the range of 0.02-0.26 μg (r = 0.999 6). The average recovery was 97.8% with a RSD of 1.7% (n = 9). The osthole was linear over the range of 0.04-0.39 μg (r = 0.999 9). The average recovery was 98.0% with a RSD of 0.9% (n = 9). Conclusion The established method is simple, accurate and reproducible, and can be used as the quality control of the two components in the preparation.
