目的观察层黏连蛋白(LN)在小鼠肾发育过程中的表达规律及层黏连蛋白与细胞极性的关系。方法用免疫组织化学链霉菌素亲和生物素-过氧化物酶连接法测定不同胚龄和日龄小鼠肾层黏连蛋白的表达。结果层黏连蛋白在肾组织中的表达随着胚龄增大和肾成熟程度而增加。在生后肾组织中的单个细胞、多个细胞无腔形成和多个细胞有腔形成时均有表达。在肾发育过程中层黏连蛋白表达于输尿管芽周围、肾小体基膜、肾小管基膜、肾小囊基膜和肾间质,且肾小管基膜着色早于肾小体基膜。结论在肾发生和成熟过程中,层黏连蛋白参与细胞极性的形成,层黏连蛋白的分布具有一定的时空顺序,对基膜的发生、成熟起重要作用。
Objective To observe the expression of laminin (LN) during mouse kidney development and the relationship between laminin and cell polarity. Methods Immunohistochemical streptavidin-biotin-peroxidase ligation assay was used to determine the expression of laminin in different age and day-old mice. Results The expression of laminin in renal tissues increased with the increase of embryonic age and the degree of renal maturation. Single cells in the kidney tissue after birth, multiple cells without cavity formation and multiple cell cavity formation are expressed. In the process of renal development, laminin expression in the ureter around the bud, the basement membrane of the renal tubule, the basement membrane of the renal tubule, the basilar membrane of the renal capsule and the renal interstitium, and tubular basement membrane color earlier than the basement membrane of the renal body. Conclusions Laminin is involved in the formation of cell polarity and the distribution of laminin in the process of kidney development and maturation. It has a certain spatial and temporal order and plays an important role in the occurrence and maturation of basement membrane.
