目的观察动脉粥样硬化大鼠高迁移率族蛋白1(HMGB-1)的蛋白及基因表达变化,以揭示与炎症相关的动脉粥样硬化发病机制。方法免疫组织化学染色法观察大鼠主动脉血管动脉粥样硬化斑块内HMGB-1的蛋白表达情况,RT-PCR法检测大鼠主动脉血管HMGB-1的基因表达变化。结果正常血管壁内存在HMGB-1表达,位于血管内皮细胞、平滑肌细胞和成纤维细胞的细胞核内;动脉粥样硬化斑块内HMGB-1分布不均匀,炎症细胞浸润区和坏死区周边基质内表达明显增高,炎症细胞浸润区内以单核-巨噬细胞胞核、胞浆和间质内表达为主,后者成条带状分布于坏死区边缘。动脉粥样硬化斑块内HMGB-1水平均明显升高,与正常血管壁比较差异有显著性(P<0.01)。结论动脉粥样硬化大鼠主动脉血管HMGB-1的蛋白及基因表达均明显增强。
Objective To investigate the protein and gene expression of high mobility group box 1 (HMGB-1) in atherosclerotic rats to reveal the pathogenesis of atherosclerosis associated with inflammation. Methods Immunohistochemical staining was used to observe the protein expression of HMGB-1 in aortic vascular atherosclerotic plaque in rats. RT-PCR was used to detect the gene expression of HMGB-1 in aorta of rats. Results The expression of HMGB-1 in the normal vessel wall was located in the nucleus of vascular endothelial cells, smooth muscle cells and fibroblasts. The distribution of HMGB-1 in atherosclerotic plaque was uneven, the infiltration of inflammatory cells and the surrounding matrix in necrotic area The expression was significantly increased, the infiltration of inflammatory cells to monocytes - macrophages in the nucleus, cytoplasm and interstitial expression of the main, which is distributed in the band edge of the necrosis area. The level of HMGB-1 in atherosclerotic plaque was significantly higher than that in normal vessel (P <0.01). Conclusion The protein and gene expression of HMGB-1 in the aorta of atherosclerotic rats were significantly increased.
