目的 通过研究重组人白介素11(rhIL-11)对正常IEC-6细胞增殖、细胞周期及其IL -1受体表达的影响,探讨IL -1对 小肠上皮细胞的调控作用以及相关机制。方法 利用MTT法检测rhIL -1对正常IEC-6细胞增殖的影响,通过流式细胞仪检测细 胞周期的变化,并采用Westernblot方法检测IEC-6细胞表面IL-11受体α链的表达情况。结果 rhIL-11可引起正常IEC-6细胞发 生增殖抑制和明显的G0/G1期阻滞,并诱导细胞表面IL 11受体α链表达增加。结论 rhIL-11可能通过与IEC-6细胞表面特异性受 体结合,从而启动胞浆内信号转导通路,引起细胞效应。
Objective To investigate the effect of rhIL-11 on proliferation, cell cycle and IL-1 receptor expression in normal IEC-6 cells and to explore the regulatory mechanism of IL-1 on small intestine epithelial cells. Methods The effect of rhIL-1 on the proliferation of IEC-6 cells was detected by MTT assay. The cell cycle was detected by flow cytometry. The expression of IL-11 receptor α chain on IEC-6 cells was detected by Western blot. Results rhIL-11 induced normal proliferation of IEC-6 cells and marked arrest of G0 / G1 phase, and induced an increase of IL-11 receptor α chain expression on the cell surface. Conclusion rhIL-11 may bind to the surface-specific receptors of IEC-6 cells, which may activate cytoplasmic signal transduction pathway and cause cellular effects.
