背景:急性脑梗死后脑组织内钙离子(Ca2+)、兴奋性氨基酸(excitatoryaminoacids,EAA)和血浆内皮素含量的升高及脑组织内镁离子(Mg2+)含量的降低都会加重脑组织的损害。给予亚低温干预后,观察急性脑梗死大鼠脑组织Ca2+和Mg2+及EAA及血浆内皮素的变化,了解亚低温的脑保护作用。目的:探讨亚低温对大鼠急性脑梗死后脑组织内Ca2+及Mg2+和EAA、血浆内皮素变化的影响及意义。设计:随机对照实验研究。单位:武汉大学人民医院神经科。材料:实验于2000-01/2000-12在武汉大学人民医院神经科实验室完成。将48只SD大鼠随机分为亚低温组及对照组,各组再分为4个亚组,每亚组6只。干预:应用改良线拴法制备大鼠大脑中动脉梗死模型;亚低温组大鼠给予亚低温治疗,而对照组不作亚低温处理。分别于缺血后1,2,4,8h处死。主要观察指标:检测各个时段大鼠缺血区脑组织内Ca2+,Mg2+,EAA及血浆内皮素含量。结果:亚低温组大鼠缺血区脑组织内Ca2+,Mg2+,EAA及血浆内皮素含量随着缺血时间的延长仅轻度升高,Mg2+含量的下降也不明显,与对照组同时段比较有显著性差异(t=62.36~135.63,P<0.01)。结论:亚低温能明显阻止实验动物脑缺血后缺血区脑组织内损伤性因子Ca2+,EAA和血浆内皮素含量的增高及保护性因子Mg2+的下降,具有脑保护作用。
BACKGROUND: Increased levels of intracellular Ca2 +, excitatory amino acids (EAA) and plasma endothelin and decreased levels of magnesium (Mg2 +) in brain tissue after acute cerebral infarction may aggravate brain damage. After mild hypothermia intervention, the changes of Ca2 +, Mg2 +, EAA and plasma endothelin in the brain of acute cerebral infarction rats were observed to understand the protective effects of mild hypothermia. Objective: To investigate the effects of mild hypothermia on changes of Ca2 +, Mg2 +, EAA and plasma endothelin in brain tissue after acute cerebral infarction in rats. Design: Randomized controlled trial. Unit: Department of Neurology, Wuhan University People’s Hospital. MATERIALS: Experiments were performed at the neurology laboratory of Wuhan University People’s Hospital from January 2000 to December 2000. Forty-eight SD rats were randomly divided into mild hypothermia group and control group. Each group was further divided into 4 subgroups, 6 in each subgroup. Intervention: The rat model of middle cerebral artery occlusion was established by modified tethering method. The rats in mild hypothermia group were treated with mild hypothermia, while those in control group were not treated with mild hypothermia. Were killed at 1, 2, 4, 8h after ischemia respectively. MAIN OUTCOME MEASURES: The contents of Ca2 +, Mg2 +, EAA and plasma endothelin in the ischemic brain tissue were detected at different time points. Results: The contents of Ca2 +, Mg2 +, EAA and plasma endothelin in the ischemic brain tissue of mild hypothermia group increased only slightly with the prolongation of ischemia time, and the decrease of Mg2 + content was not obvious. Compared with the control group There was a significant difference (t = 62.36 ~ 135.63, P <0.01). CONCLUSION: Mild hypothermia can obviously prevent the increase of the content of injury factors Ca2 +, EAA and plasma endothelin and the decrease of protective factor Mg2 + in the ischemic brain tissue of experimental animals after cerebral ischemia.
