目的:对大孔树脂富集纯化荆芥穗总黄酮的工艺条件及参数进行优化。方法:先以总黄酮比吸附量和解吸率为指标,通过静态吸附与解吸试验对9种树脂型号进行筛选,再通过动态吸附与解吸试验对富集纯化工艺参数进行全面优化。结果:HP-20树脂对荆芥穗总黄酮的吸附与解吸性能较好;确定最佳吸附条件为径高比1∶6,样品溶液pH值为6.0,浓度0.4 g(生药)/mL,吸附流速2 BV/h和最佳吸附量133.76 mg(总黄酮)/g(干树脂);最佳解吸条件为80%乙醇洗脱,洗脱流速4 BV/h和溶剂用量3 BV。结论:HP-20树脂可有效地对荆芥穗总黄酮进行富集纯化。
OBJECTIVE: To optimize the process conditions and parameters for the purification of total flavonoids in Nepeta by macroporous resin. Methods: The total flavonoids than the amount of adsorption and desorption rate as an indicator, through the static adsorption and desorption tests on the 9 kinds of resin types were screened, and then by dynamic adsorption and desorption of enrichment purification process parameters to fully optimize. Results: The adsorption and desorption properties of HP-20 resin on the total flavonoids of Nepeta were good. The optimum adsorption conditions were as follows: diameter-height ratio of 1: 6, pH value of the sample solution of 6.0, concentration of 0.4 g Flow rate of 2 BV / h and the best adsorption amount of 133.76 mg (total flavonoids) / g (dry resin); the best desorption conditions were 80% ethanol elution flow rate of 4 BV / h and the amount of solvent 3 BV. Conclusion: HP-20 resin can effectively enrich and purify the total flavonoids of Nepeta.
