目的 从细胞及组织水平检测血小板源性生长因子受体 - β(PDGFR- β)在正常皮肤及瘢痕疙瘩中的表达及分布 ,探讨 PDGFR- β在瘢痕疙瘩形成中的生物学作用。方法 1应用免疫组织化学技术检测 PDGFR-β在正常皮肤和瘢痕疙瘩组织中的分布 ;2应用免疫荧光、流式细胞仪检测正常皮肤和瘢痕疙瘩成纤维细胞群体的PDGFR- β的表达 ;3应用免疫荧光、激光共聚焦显微镜检测 PDGFR- β的亚细胞分布。结果 1免疫组织化学 正常皮肤与瘢痕疙瘩组织中 PDGFR- β表达基本相同 ;2流式细胞仪检测 正常皮肤成纤维细胞 PDGFR- β的表达明显高于瘢痕疙瘩成纤维细胞 ,有统计学意义 (P<0 .0 1) ;3激光共聚焦显微镜检测 在正常皮肤成纤维细胞中PDGFR- β主要集中于细胞核膜及核内 ,瘢痕疙瘩成纤维细胞中 PDGFR- β主要集中于细胞膜。结论 1体外培养的成纤维细胞和组织中成纤维细胞 PDGFR- β有差异 ;2正常皮肤成纤维细胞 PDGFR- β表达高于瘢痕疙瘩成纤维细胞 ;3正常皮肤成纤维细胞与瘢痕疙瘩成纤维细胞中 PDGFR- β亚细胞分布不同 ,这可能是瘢痕疙瘩形成的重要因素。
Objective To detect the expression and distribution of platelet - derived growth factor receptor - β (PDGFR - β) in normal skin and keloid from the cellular and tissue level and to explore the biological role of PDGFR - β in the formation of keloid. Method 1 Immunohistochemistry was used to detect the distribution of PDGFR-β in normal skin and keloid tissues; 2 The expression of PDGFR-β in normal skin and keloid fibroblasts was detected by immunofluorescence and flow cytometry; 3 Immunofluorescence and laser confocal microscopy were used to detect the subcellular distribution of PDGFR-β. Results1 The expression of PDGFR- β in normal skin and keloid tissues was similar in immunohistochemistry. 2 Flow cytometry showed that the expression of PDGFR- β in normal skin fibroblasts was significantly higher than that in keloid fibroblasts (P <0.01). 3 Confocal laser scanning microscopy showed that PDGFR-β mainly localized in the nucleus and nucleus in normal skin fibroblasts. The majority of PDGFR-β in keloid fibroblasts were mainly concentrated in the cell membrane. Conclusion 1 PDGFR-β fibroblasts cultured in vitro and in tissue differentiated; 2 normal skin fibroblasts PDGFR-β expression was higher than keloid fibroblasts; 3 normal skin fibroblasts and keloid fibroblasts In PDGFR-β subcellular distribution, which may be an important factor in the formation of keloids.
