扩增了光敏核不育系农垦58S 及从它衍生出来的 5 个两用核不育系叶绿体 DNA 的 ORF(openreadingframe)100、ORF29-TrnCGCA、rps16(ribosomalproteins16)基因内含子和TrnTUGU-TrnLUAA(tRNA-Thr(UGU)-tRNA-Leu(UAA))等4个片段,并测定了其序列。研究结果表明,粳型光敏核不系农垦58S 的叶绿体为粳型。农垦58S 衍生的核不育系中,粳型核不育系7001S以及3个籼型核不育系1103S、培矮64S 和广占63S 的叶绿体DNA 为粳型,与选育者提供的细胞质系谱一致。籼型核不育系W6154S 的叶绿体DNA 为籼型,与选育者提供的细胞质系谱不一致,推断选育者在选育过程中更换过细胞质(曾用不育系作过父本)。5 个粳型叶绿体DNA 的rps16 基因内含子和TrnTUGU-TrnLUAA 间区的序列相互之间有1 ̄2个单核苷酸的变异。
The ORF (open reading frame) 100, ORF29-TrnCGCA, rps16 (ribosomal protein 16) gene introns and TrnTUGU-TrnLUAA (+) of chloroplast DNA of Nongken 58S and five dual- tRNA-Thr (UGU) -tRNA-Leu (UAA)), and their sequences were determined. The results showed that the chloroplasts of japonica type photoperiod nuclear line No Nong Ken 58S were japonica. Among Nongken 58S-derived male sterile lines, the japonica CMS line 7001S and the three indica CMS lines 1103S, Peiai 64S and Guanghan 63S were chloroplast DNAs, which were similar to the cytoplasmic lines Consistent. The chloroplast DNA of indica type CMS line W6154S was indica type, which was different from that of the cytoplasmic line provided by the breeder. It was deduced that the breeder replaced the cytoplasm during the breeding process. The sequence of intron of rps16 gene and the region of TrnTUGU-TrnLUAA among 5 japonica chloroplast DNAs had 1 ~ 2 single nucleotide variations among them.
