目的分别用血清脑心浸液和β-环糊精脑心浸液两种培养基培养幽门螺杆菌,观察幽门螺杆菌在两种培养基中的生长规律。方法将幽门螺杆菌分别接种于血清脑心浸液和β-环糊精脑心浸液中,置37℃微需氧环境以120r/min震摇培养,于不同时间点以稀释平板计数法计数活菌数量,制作生长曲线。结果幽门螺杆菌在血清脑心浸液和β-环糊精脑心浸液中的迟缓期均为10h,对数期分别为45h和40h,对数中期时比生长速率分别为0.058/h和0.043/h,稳定期分别为15h和20h,最高浓度分别为2.067×10~8 CFU/ml和1.378×10~8 CFU/ml。结论幽门螺杆菌在血清脑心浸液和β-环糊精脑心浸液中均生长良好,其中在血清脑心浸液中最高生长密度更大,对数期维持时间较长。
OBJECTIVE To culture Helicobacter pylori with serum brain heart infusion and β-cyclodextrin brain-heart infusion, respectively, and observe the growth of Helicobacter pylori in the two culture media. Methods Helicobacter pylori were inoculated into the brain stem and brain infusions of β-cyclodextrin and brain. The cells were shaken at 120r / min in aerobic environment at 37 ℃ and counted by dilution plate counting at different time points The number of viable bacteria, production growth curve. Results Helicobacter pylori in retina and β-cyclodextrin brain stem and heart infusion were lag phase 10h, logarithmic phase were 45h and 40h, the logarithmic metaphase than the growth rate were 0.058 / h and 0.043 / h, the stationary phase was 15h and 20h, the highest concentrations were 2.067 × 10 ~ 8 CFU / ml and 1.378 × 10 ~ 8 CFU / ml, respectively. Conclusions Helicobacter pylori can grow well in both brain and heart extracts of brain and heart, and the highest growth density in brain and heart extracts of serum is higher and the logarithmic period is longer.
