目的观察养心开郁片对小鼠急性抑郁模型的影响。方法应用小鼠悬尾模型、游泳不动时间模型、对小鼠自主活动的影响、五-羟色胺酸诱导的小鼠甩头行为模型四种方法,分为5组。养心开郁片小、中、大剂量组分别给予每日310 mg·kg-1、620 mg·kg-1、1240 mg·kg-1(以片重计)。前3种方法,以路优泰为阳性对照,每日剂量150 mg·kg-1,第4种方法以盐酸氟西汀为阳性对照,剂量10 mg·kg-1。连续14 d。结果 1、养心开郁片大剂量组可使小鼠悬尾不动时间显著减少;游泳不动时间显著减少。2、养心开郁片小、中、大剂量组对小鼠自主活动无明显影响。3、养心开郁片大剂量组小鼠甩头潜伏期明显缩短,甩头次数显著增加。结论养心开郁片显示出明显的抗抑郁作用,其作用机制与影响5-HT的代谢有关,对中枢神经系统无明显兴奋或抑制作用。
Objective To observe the effects of Yangxin Kaiyu tablets on acute depression model in mice. Methods Four methods of mice tail suspension model, swimming time-invariant model, their effect on autonomic activity and pentylenetetrazol-induced mouse head-shaving behavior model were divided into five groups. Yang Xin Kai Yu tablets small, medium and large dose groups were given daily 310 mg · kg-1,620 mg · kg-1,1240 mg · kg-1 (by weight). The first three methods to Lu You-tao as a positive control, a daily dose of 150 mg · kg-1, the fourth method of fluoxetine hydrochloride as a positive control, a dose of 10 mg · kg-1. For 14 days. Results 1, Yang Xin Kai Yu tablets high-dose group can make the mouse tail time is significantly reduced; swimming time is significantly reduced. 2, Yang Xin Kai Yu film small, medium and large dose group had no significant effect on the autonomy of mice. 3, Yang Xin Kai Yu tablets high-dose group mice shook head significantly shorter incubation period, shampooing times increased significantly. Conclusion Yang Xin Kai Yu film showed a significant antidepressant effect, its mechanism of action and the impact of 5-HT metabolism, the central nervous system without significant excitement or inhibition.
