目的:建立高效液相色谱法测定痫乐散中苯巴比妥的含量。方法:采用Diamonsil(钻石)C18色谱柱(5μm,250mm×4.6mm);以甲醇-磷酸盐缓冲液(磷酸二氢钾6.8g,加三乙胺4mL,加水至1000mL)(47:53)为流动相;流速:1.0mL/min;检测波长:215nm;进样量:20μL。结果:苯巴比妥线性范围为6~60μg/mL,r=0.9999;平均回收率为99.9%,RSD=0.52%,n=9。结论:该方法简便准确,专属性好,可以作为痫乐散中苯巴比妥含量的测定方法。
Objective: To establish a method for the determination of phenobarbital in epilepsy powder by high performance liquid chromatography. METHODS: Diamonsil (C18) column (5μm, 250mm × 4.6mm) was used. The column was washed with methanol-phosphate buffer (potassium dihydrogen phosphate 6.8g, triethylamine 4mL and water 1000mL) Mobile phase; Flow rate: 1.0 mL / min; Detection wavelength: 215 nm; Injection volume: 20 μL. Results: The linear range of phenobarbital was 6 ~ 60μg / mL, r = 0.9999; the average recovery was 99.9%, RSD = 0.52%, n = 9.
