目的:观察冠心宁注射液对大鼠心肌缺血再灌注损伤梗死面积以及心肌热休克蛋白70(HSP70)表达的影响,探讨冠心宁的心肌缺血再灌注损伤的保护作用。方法:45只雄性SD大鼠随机分成3组(每组n=15):对照组(Ⅰ组)、缺血再灌注组(Ⅱ组)、冠心宁治疗组(Ⅲ组),建立在体心肌缺血再灌注动物模型。每组5只动物再灌注40min后测定梗死面积,测定超氧化物歧化酶(SOD)活性和心肌丙二醛(MDA)含量;每组另外10只动物再灌注120min后观察心肌细胞HSP70蛋白表达。结果:再灌注前注射冠心宁注射液显著降低了再灌注心肌的梗死面积、MDA水平(P<0.01),增强了SOD活性(P<0.01)和HSP70的表达(P<0.01)。结论:冠心宁注射液可通过抗氧化作用、增加HSP70的表达发挥抗心肌缺血再灌注损伤,降低心肌梗死面积,对大鼠心肌有明显的保护作用。
Objective: To observe the effect of Guanxinning injection on infarct size and heat shock protein 70 (HSP70) expression in rats with myocardial ischemia-reperfusion injury, and to explore the protective effect of Guanxinning on myocardial ischemia-reperfusion injury. Methods: Forty-five male SD rats were randomly divided into 3 groups (n = 15 in each group): control group (group I), ischemia-reperfusion group (group II), Guanxinning treatment group (group III), established in vivo Myocardial ischemia-reperfusion animal model. The infarct size was measured after 5 minutes of reperfusion in each group. Superoxide dismutase (SOD) activity and myocardial malondialdehyde (MDA) content were measured. After 10 minutes of reperfusion, the expression of HSP70 protein was also observed in each group. Results: Injection of Guanxinning injection before reperfusion significantly reduced the infarct size and MDA level (P<0.01), and enhanced the activity of SOD (P<0.01) and HSP70 (P<0.01). Conclusion: Guanxinning injection can exert anti-myocardial ischemia reperfusion injury by reducing the expression of HSP70, reducing the infarct size of myocardium, and has obvious protective effect on rat myocardium.
