目的定量观察和分析胸水涂片中腺癌细胞的细胞浆、细胞核和全细胞的色度学特征,筛选对判别ACC和反应性间皮细胞有参考价值的色度学参数。方法胸水离心后取沉淀物涂片、然后进行巴氏染色;在数码显微镜下拍摄图像,用计算机图像分析技术测量其中的764个ACC(44例)和709个RMC(39例)的色度学参数,包括细胞浆、细胞核和全细胞的红、绿、蓝三基色,并对测试结果进行判别分析,比较基于三种色度学参数所建立函数的判别效果。结果与RMC相比,ACC的细胞浆、细胞核和全细胞红、绿、蓝三基色明显减小(P<0.05)。基于以上三种色度学参数所建立函数的判别符合率分别为68.7%、59.4%和74.7%,其中基于全细胞色度学参数所建立的函数判别符合率明显高于基于细胞浆和细胞核色度学参数所建立函数的(P<0.05)。结论在胸水ACC的定量判别分析中,全细胞的色度学参数比细胞浆和细胞核可能更具有参考价值。
OBJECTIVE: To quantitatively observe and analyze the chromatographic features of cytoplasm, nucleus and whole cells of adenocarcinoma cells in pleural effusion, and to screen the chromosomal parameters that can be used as a reference for discriminating ACC and reactive mesothelial cells. Methods Pleural effusion was centrifuged and smear sediment, followed by pasteurization. Images were taken under a digital microscope and the colorimetry of 764 ACCs (44 cases) and 709 RMCs (39 cases) was measured by computer image analysis Parameters, including the cytoplasm, nucleus and whole cells of the red, green and blue primary colors, and the test results were discriminant analysis, the comparison based on the three colorimetric function established discriminant effect. Results Compared with RMC, the primary, mitochondrial, nuclear and whole cell red, green and blue of ACC decreased significantly (P <0.05). The coincidence rates of the functions established based on the three colorimetric parameters were 68.7%, 59.4% and 74.7% respectively. The coincidence rate of the discriminant function based on the whole-cell colorimetric parameters was significantly higher than that based on the cytoplasm and the nucleus color The degree of learning parameters established by function (P <0.05). Conclusions In quantitative analysis of pleural effusion ACC, whole cell chromatographic parameters may be more valuable than cytoplasm and nucleus.
